Extraction of Phytochemicals from Spinach (Spinacia Oleracea) and their Antioxidant Activity /Anam Ishfaque

Summary: The plant material (Spinacia oleracea) was collected from the local agricultural field near Lahore. Collected material was washed to remove debris, cut into small pieces and then it was shade dried. Constituents of Spinacia oleracea were extracted by soaking dried material into methanol. The purpose of this study was to investigate phytochemicals present in Spinacia oleracea and determination of their antioxidant activity. Preliminary phytochemical screening revealed the presence of secondary metabolites such as alkaloids, steroids, saponins, tannins and phenolic compounds which have been reported for many biological activities. UV-visible absorption spectra of chlorophyll presented maximum absorption at 406nm. Total phenolic content (TPC) was determined by using folin-ciocalteu reagent and reading was taken at 765nm by UV-Visible spectrophotometer. Gallic acid was used as standard and the sample showed absorbance of 0.002 and 0.083% TPC. Free radical scavenging activity was measured by DPPH (2,2-Diphenylpicrylhydrazyl radical) by taking 10, 20, 30, 40 and 50 μL of the samples. The absorbance was taken at ƛ517nm. It was found that antioxidant activity was concentration dependent, as the concentration increases the antioxidant activity also increases. Maximum activity 76.92% was obtained with 50 μL concentration. Phenolic compounds which are considered as potent antioxidants prevent oxidation of biomolecules and thus are involved in reduction of tissue damage.
