Summary, etc Antibiotics are the drugs or medicines that are obtained from livings organisms and used to treat bacterial or fungal infections. Long term use of antibiotics against a certain microorganism lead to the adaptation of microorganism against that specific antibiotic which is commonly called as antibiotic resistance. Antimicrobial resistance is the capability of that organism that can be seen under the microscope. It is their prime quality that it opposes the impact of an antitoxin. Bacteriophages are infectious agents which attack on bacteria cells, on lytic phages, disturb bacteria digestion, and destruct them. Lytic Bacteriophages are used as phage therapy to cure pathogenic maladies. The Research aim was to characterize the bacteriophage specific to bacterium Pseudomonas aeruginosa and to study bacteriostatic efficiency (reduction capacity) of phages specific to the PA-1. For characterization of bacteriophage IQL against clinically isolated Pseudomonas aeruginosa strain sample was collected from different areas of Lahore but the required sample was finalized from Iqbal Town, Lahore. The sample was processed for bacteriophage enrichment by standard protocols. Then, Bacteriophage was characterized by temperature stability, pH stability, storage stability and reduction assay to reduce the bacterial growth and one step growth curve to calculate the burst time and then its genome was characterized. The Bacteriophage IQL has a narrow spectrum host range. It was effective against 20/38 P. aeruginosa, 1/3 Klebsiella pneumoniae, ½ S. marcescens strains and have no lytic activity against Acinetobacter baumannii, Escherichia coli, Burkholdera cepacia, and enterobacter strains under study. The titer of the Bacteriophage IQP was counted 9.4×1021 PFUs/ml. Bacteriophage IQL was stable at pHysiological pH range of 5-9. The best activity is observed at alkaline pH of 9, whereas it showed slight decrease in the PFUs at pH below 9. For survey of temperature impacts on bacteriophage suitability, different temperatures were used to ascertain titer for one hour phages cure. The bacteriophage stabled on 25, 37 and 45 oC. After curing at 60 oC on phages noticeable decrease seen in titer. On 4 oC, 25 oC, and 37 oC temperature titer reduction was not seen following storage of one month in phage IQL. Slight decrease in the titer was seen at -20 and - 80oC for one month. IQL 2 bacteriophage reduces the bacterial growth completely till 12 hours at MOI 10.
